Jason Isaacs has said he’d enjoy bringing the Grand Inquisitor to live action, although mostly because of his cool spinny Inquisitor lightsaber. He looks enough like the character for the part, and obviously sounds it - I’d love to see this happen someday.
I like stories about the relationship of hunter and hunted, but personally not sure about the idea of an Inquisitor falling in love with the Jedi they’re hunting. A lot of outlandish ideas in Star Wars work for me, but this one seems a stretch and hard to accomplish. Also, more Jedi on the run in the ESB/ROTJ time period lessens the significance of Luke’s journey (Cal Kestis for example should, imo, die a heroic death sometime before A New Hope)
I think it’s difficult to accomplish really interesting stories around the Inquisitors. I haven’t yet played Fallen Order, but it looks like the story of an Inquisitor and a Jedi who knew each other before the purge reconciling has already been done, Inquisitors like Fifth Brother and Seventh Sister played their part in Rebels - as recurring side-villains so Vader could be used sparingly - just fine, but to my knowledge they’re not incredibly interesting characters by themselves.
