I believe it's Episode CDXX, "Revenge of the Ganja",
the scene when Luke takes a huge hit.
...Sorry, I don't believe Yoda says this in any of the movies.

I'm sure whoever you got this quote from, purposely distorted it for humorous effect.
The scene is from "The Empire Strikes Back", here's a direct quote from the scene in Yoda's hut.
Luke - I am ready! Ben! I... I can be a Jedi, Ben, tell him I'm rea- [BONKS HEAD]
Yoda - Ready are you? What know you of ready? For eight hundred years have I trained Jedi...
