Probably when Lando says...
"We're on our way, Red group, Gold group, all fighters follow me. Ha ha ha, I told you they'd do it!"
...followed by an EPIC music sting and heroic FX shot :-)
It's all I can do to not jump up and down with excitement when he says that line, no matter how many viewings and years have since I first heard it.
Not the most original line (You could slot it into a WWII movie without trace, as you could with many SW lines) but it's the way Billy Dee Williams delivers it.

Original Boba Fett's line "As you wish" is a strong contender. There are so many layers to those 3 words the way it is delivered.

"Are you brain dead?" This is my favorite Star Wars quote for many reasons. First, I love this quote because of how memorable and popular it has become. Think about it. When was the last time you went out in public and NOT seen someone with an "Are you brain dead?" T- shirt? There are so many more quotes I love too, like "There's always a bigger fish" and "Yipee!"
