Luke had a computer on board his X-wing that interpreted R2. You can see him looking at it when he’s talking to R2 on the flight to Dagobah. As for other times he speaks to R2, you can imagine he’s verbalizing non-verbal communication. There are probably intonations in the tone of R2’s beeps that can kind of intimate what the droid might be trying to convey. The same way you might talk to your cat or dog but neither really knows what the other is trying to say.
